Проверить предположение о том, что в митохондриях существуют отдельные пулы миторибосом, специализирующихся на трансляции отдельных мРНК
Mitochondria are important organelles of almost all eukaryotic cells, performing many functions such as energy biosynthesis in the form of ATP, the formation of iron-sulfur clusters, participation in apoptosis, fatty acid metabolism, etc. According to the generally accepted endosymbiotic theory, mitochondria descended from the ancestor prokaryote similar to alphaproteobacteria, and over billions of years of evolution almost completely lost the original genome. Most of the genetic information has been transferred to the nucleus or lost. More than 99% of the mitochondrial proteins of modern eukaryotes are encoded in the nucleus, synthesized in the cytosol, and then imported into mitochondria. Nevertheless, mitochondria retained their own genome, carrying genes of about 10 proteins - components of complexes of the oxidative phosphorylation chain, a minimal set of tRNAs, ribosomal RNAs, as well as transcription and protein biosynthesis apparatus. Studies of mitochondrial translation have attracted the attention of many scientists worldwide, since any disorders in this process lead to mitochondrial dysfunction and the development of many serious diseases, such as neuropathies, muscle dystrophies, neurodegenerative diseases, etc. It was previously believed that protein biosynthesis in mitochondria was generally organized according to the bacterial type, however, studies of recent decades have shown that mitochondrial translation, although it has similarities with bacterial, has several significant differences. First, mitochondrial ribosomes, unlike bacterial ones, have a significantly higher protein content relative to RNA, which, most likely, is associated with the evolutionary replacement of the functions of individual sections of ribosomal RNAs with proteins. In addition, according to cryoelectronic microscopy, 5S rRNA is absent in mammalian mitoribosomes, and valine (in humans) or phenylalanine (in porcine) tRNA takes its place. In addition to differences in the structures of the ribosomes themselves, protein biosynthesis in bacteria and mitochondria differs significantly in the context of the regulation of this process. So, in all the studied mitochondrial translation apparatus there is no initiation factor 1 that is absolutely necessary in bacteria, and its functions, at least in yeast and mammals, are performed by a small domain of initiation factor 2. In addition, as was shown in our laboratory, the absence of a third initiation factor translation (IF3), which is necessary for initiating protein biosynthesis in bacteria, does not completely stop translation in yeast mitochondria but is a cause of imbalance in protein synthesis. We obtained similar results for human mitochondria, while in the absence of mitochondrial IF3, the synthesis of cytochrome c oxidase components is reduced in yeast and one of the components of ATP synthetase in human mitochondria. An important factor determining the need for fine regulation of mitochondrial translation is the necessity for its coordination with the biosynthesis of mitochondrial proteins in the cytosol for stoichiometrically correct assembly of respiratory chain complexes. In yeast mitochondria, specific regulation of the translation of individual mitochondrial mRNAs is provided by a set of special proteins — translational activators that interact with extended 5’-UTRs and determine the efficiency of biosynthesis of mitochondria-encoded polypeptides. At the same time, mammalian mitochondrial mRNAs are practically devoid of untranslated regions; moreover, only one true translational activator, TACO1 protein, which regulates the translation of COI mRNA, has been identified so far. The principles of regulation of translation of other mRNAs in mammalian mitochondria remain almost unexplored. One of the concepts proposed in the scientific community for the regulation of mitochondrial translation in mammals is the assumption that there are separate pools of mitoribosomes in mitochondria specializing in the translation of individual mRNAs. Mitoribosomes from different pools have a similar structure, however, they differ from each other by the presence of certain protein factors that determine the specificity of the translation initiation of one or another mRNA. In the framework of the proposed project, we plan to describe the mechanisms of regulation of translation in mammalian mitochondria. To do this, we intend to use an integrated approach using bioinformatics, molecular biology and biochemistry methods. We will define the set of candidate mitoribosomal proteins capable of differentiating the translation initiation of certain mRNAs, after which we will evaluate the effect of knockdown or knockout of the corresponding genes on mitochondrial translation and functional activity of mitochondria. In addition, we plan to assess the possible role of initiation factors (mtIF2 and mtIF3) in discrimination against the specificity of translation of individual mitochondrial mRNAs.
В результате выполнения проекта будут получены принципиально новые научные данные о механизмах регуляции трансляции в митохондриях клеток млекопитающих. Мы планируем оценить влияние нокдауна и/или нокаута как минимум 6 генов белков, ассоциированных с митохондриальными рибосомами человека, на трансляцию митохондриальных мРНК. Мы предполагаем, что подавление экспрессии или делеция генов таких белков приведет к снижению эффективности биосинтеза отдельных митохондриально закодированных белков, т.е., к разбалансировке митохондриальной трансляции, что будет свидетельствовать о функции таких белков как специфических факторов трансляции отдельных мРНК. Поскольку скоординированность цитозольной трансляции и биосинтеза белка в митохондриях является необходимым условием корректной сборки комплексов и суперкомплексов комплексов цепи окислительного фосфорилирования внутренней мембраны митохондрий, мы планируем оценить влияние полученных нами делеций (или клеток с пониженной экспрессией соответствующих генов) на сборку и активность отдельных комплексов электрон-транспортной цепи и их способность к образованию суперкомплексов. Помимо этого, нами будет проанализирован уровень потребления клетками кислорода в условиях подавления экспрессии кодирующих указанные белки генов. В случае успеха этой части проекта мы планируем оценить количественное соотношение найденных белков-регуляторов и коровых белков миторибосом для подтверждения или опровержения концепции о существовании отдельных пулов миторибосом, специализирующихся на трансляции различных мРНК. В результате выполнения проекта мы планируем получить следующие результаты: - список белков, ассоциированных с миторибосомами, являющихся потенциальными регуляторами трансляции отдельных мРНК; - как минимум 6 линий клеток человека, в которых гены упомянутых белков функционально делетированы с использованием технологии редактирования генома CRISPR/Cas9, или же экспрессия этих генов подавлена с помощью РНК-интерференции; - расчет эффективности трансляции всех 13 митохондриально закодированных белков в вышеупомянутых линиях клеток, количественные данные по активности комплексов цепи окислительного фосфорилирования, расчет эффективности и корректности сборки комплексов электрон-транспортной цепи и их способность к организации в суперкомплексы; - расчет стехиометрических соотношений выбранных нами факторов и коровых белков митохондриальных рибосом человека. Результаты выполнения проекта будут востребованы мировым научным сообществом, поскольку в последние годы интерес к исследованиям митохондриальной трансляции лавинообразно увеличивается по причинам все увеличивающегося количества сообщений о связи нарушений в митохондриальной трансляции с широким спектром тяжелых заболеваний и существенными достижениями в структурной биологии миторибосом. Результаты проекта, помимо важности в понимании механизмов тонкой регуляции трансляции в митохондриях, будут способствовать созданию системы митохондриальной трансляции in vitro, необходимой как для изучения механизмов трансляции в митохондриях, так и для тестирования новых лекарственных препаратов.

Результаты этапа: План работ выполнен в полном объеме. Получены линии клеток человека с делециями в генах CHCHD1, PTCD2 и PTCD3. Для полученных линий определена эффективность митохондриальной трансляции, измерены скорости потребления кислорода, изучены активности комплексов цепи окислительного фосфорилирования, установлен стехиометрический состав суперкомплексов. Показано, что белок PTCD2 является селективным регулятором трансляции митохондриально кодируемого белка Cox3. | ||
