Cellulose and hemicelluloses are the major components of plant cell walls and the main substrates in processes of biotransformation of lignocellulosic materials. Different methods of protein engineering are used to improve biochemical properties of enzymes and increase efficiency of technological processes. Site-directed mutagenesis of N-linked glycosylation sites was used as a way to change the biochemical properties of cellobiohydrolase I from Penicillium verruculosum. A significant increase of hydrolytic activity of CBHI was demonstrated towards avicell as a model substrate and wood of aspen as a natural substrate.
